Understanding the functional role of genome maintenance pathways and the canonical DNA damage response in mammalian physiology is central to uncovering mechanisms underlying human health and disease. Our research focuses on elucidating the interplay between DNA repair mechanisms, transcriptional stress, immune signaling, and DNA damage-induced inflammatory responses. Using unique animal models carrying inborn defects in DNA repair-associated genes, in combination with molecular, cellular, and genomic approaches, we investigate how persistent genome instability drives DNA damage-driven pathologies, including chronic inflammation and age-related dysfunction.
